Investor presentation summary
12 Oct, 2026AO-252 biology and differentiation
Only clinical-stage program targeting TACC3, a selective cancer survival dependency supported by DepMap analysis across multiple tumour types.
Oral TACC3 protein-interaction inhibitor designed to cause DNA damage and replication stress, with cGAS/STING-driven immune activation alongside direct cancer-cell killing.
Differentiation includes blood-brain barrier penetration, biomarker-driven patient selection, and potential for monotherapy and combination use.
Preclinical evidence
Tumour regression reported across ovarian, TNBC, endometrial, gastric and prostate models, including brain metastases; TGI of 88–120% and partial-to-complete responses across tested indications.
No toxicities observed in 28-day GLP rat and dog studies; only 4 of 468 kinases inhibited by >65% at 1 μM, with a reported therapeutic window of ≥3–5X.
Phase I progress
Cohort 4b with twice-daily dosing showed an 80% clinical benefit rate and 20% ORR; no SAEs, median five prior therapies, and duration of response ≥6 months.
Clinical benefit and tumour reductions in ovarian and endometrial cancer emerged before targeted exposure levels; MTD had not been reached, and BID dosing increased CBR from 40% to 80%.
Lipid-based softgel formulation is active; 120 mg BID and 160 mg QD cohorts enrolled, with dosing of up to 30 patients targeted by YE 2026.
Phase I includes dose escalation and expansion in advanced solid tumours, with or without brain metastases; endpoints include safety, PK, response rate, duration of response and survival.
