Chardan's 10th Annual Healthcare Conference summary
20 Oct, 2026Clinical pipeline and milestones
LX2006 is the lead program for Friedreich’s ataxia (FA) cardiomyopathy; its registrational SUNRISE-FA 2 study began enrolling at the end of Q2.
Phase I/II data showed effects on left ventricular mass index, multiple biomarkers and neurological function measured by mFARS.
LX2006 phase III top-line data are expected in 2H 2027, followed by a planned BLA filing in H1 2028.
LX2020 targets PKP2-associated arrhythmogenic cardiomyopathy; phase I/II data showed effects on NSVT and PVCs with a favorable safety profile.
Preclinical programs include LX2021 for desmoplakin cardiomyopathy and LX2022 for hypertrophic cardiomyopathy.
PKP2 data and regulatory outlook
January data showed PVCs down 14% and NSVT down 22%; two patients with nine-month follow-up had a 65% NSVT reduction.
Q4 update is expected to include most high-dose patients with at least 12 months of follow-up, plus a regulatory update; most of seven high-dose patients are expected to reach 12 months, except one or two.
NSVT is viewed as the most clinically meaningful endpoint; a 35%-40% reduction was described as a target to exceed the 20%-30% effects associated with current drug treatments.
FDA discussions on endpoints and pivotal design are ongoing; an August RMAT designation submission included contemporary data centered substantially on NSVT.
Arrhythmia effects are expected to deepen over time, with 12 months viewed as an initial point to assess response; structural changes may take longer, and one patient showed increased RVEF.
FA strategy and study execution
LX2006 reaches peripheral nervous system components, including skeletal muscle and dorsal root ganglia, but not the brain; the Mantle portfolio adds programs intended to reach the brain and potentially complement LX2006 in FA.
CLARITY-FA serves as a feeder into SUNRISE-FA 2, with identical inclusion criteria; eligible participants may roll over if they test negative for neutralizing antibodies and their treatment site is activated.
CLARITY-FA had 19 active sites across eight countries and 22 enrolled patients; SUNRISE-FA 2 is designed for 13 treated and 13 untreated control patients.
Confirmatory approval discussions continue; potential evidence may combine cardiac endpoints and mFARS.
