Strategic thesis and portfolio

• Rationale: upstream immune signaling nodes may enable deeper, more durable remissions; portfolio targets RIPK2, SLC15A4, TNFR2 and TSLP/IL-33 across I&I diseases.

• OD-001 is an oral RIPK2 inhibitor for UC and Crohn’s disease; OD-002 is an oral SLC15A4 inhibitor for cutaneous lupus, nephropathies and B-cell-mediated disease.

• OD-003 is a TNFR2 protein agonist for atopic dermatitis, SLE and alopecia areata; TSLP/IL-33 protein OD-004 targets COPD and asthma.

OD-001 in inflammatory bowel disease

• Phase 2a UC monotherapy met the primary MMCS endpoint, with a -2.6 change from baseline (p<0.001); 27% of 49 participants achieved clinical remission at week 12 versus a 10% historic placebo benchmark.

• Clinical remission was 25% in advanced-therapy-experienced patients; the presentation also reported 4 clinical remissions and 1 additional endoscopic improvement in this subgroup.

• Data were from an ongoing study and subject to change before clinical study report finalization; 20 of 45 participants in the 20 mg BID group had a TEAE, with no severe TEAEs or SAEs reported.

• Planned UC development includes a phase 2a vedolizumab combination induction readout and phase 2b monotherapy readout in H2 2027; monotherapy maintenance results are expected in 2028.

OD-002 in B-cell-mediated disease

• OD-002 selectively inhibits SLC15A4 signaling downstream of TLR7/8/9; preclinical data showed blockade of RNA- and DNA-driven signaling and pathogenic B-cell activity while sparing healthy B-cell responses.

• In lupus-prone mice, OD-002 reduced age-associated B cells by 57% and anti-dsDNA antibodies by 66%; proliferating B cells in healthy mice were spared.

• A CTA filing is planned for H2 2026, phase 1/2 initiation for H1 2027, and phase 2a readouts across multiple indications for H2 2028.
