Peritoneal metastases from various cancers have limited treatment options, with surgery as the only curative approach and high relapse rates due to residual micrometastases.
Ovarian cancer patients frequently present with peritoneal micrometastases, and up to 85% relapse after resection, with recurrence mainly in the peritoneum.
Controlling residual or reseeded disease post-surgery is crucial for improving survival and quality of life.
Product overview and mechanism
RadspherinⓇ is an alpha-emitting radiopharmaceutical (224Ra on CaCO3 microparticles) designed for local retention in the peritoneum.
Administered 1-3 days post-surgery, it delivers a high dose of radiation to micrometastases while sparing normal tissue.
Single-dose administration fits into existing clinical workflows and minimizes patient burden.
The mechanism is independent of tumor origin or mutation, enabling broad applicability across peritoneal-spreading cancers.
Clinical efficacy and safety
In ovarian cancer Phase 1, 90% of patients remained free of peritoneal recurrence at 24 months, with overall recurrence at 30%.
In colorectal cancer Phase 1/2a, 72% remained recurrence-free at 18 months, with a peritoneal recurrence rate of 28% versus 50% in literature benchmarks.
No dose-limiting toxicity observed across 68 patients; adverse events were expected and manageable.
Only three serious adverse events possibly related to RadspherinⓇ were reported.
