Jefferies Global Healthcare Conference 2026 summary
3 Jun, 2026Strategic vision and market opportunity
Focus on transforming cystic fibrosis (CF) treatment by targeting the NBD1 region of the CFTR protein, aiming to address the significant unmet need where only one-third of patients on current therapies reach normal CFTR function.
The CF market is valued at $12–13 billion and is projected to exceed $15 billion by the end of the decade, with a monopoly currently held by a single competitor.
Sionna’s approach leverages a rich scientific history, originating from Genzyme and Sanofi, and focuses on differentiated NBD1 stabilizers.
Scientific and clinical development
NBD1 stabilizers are designed to directly address the F508del mutation, present in 90% of CF patients, by stabilizing the NBD1 domain and improving protein function beyond current standards.
Preclinical data show that NBD1 monotherapy can match the efficacy of current triple combinations, and dual combinations may drive more patients to normal CFTR function.
Two lead compounds, SION-719 and SION-451, are advancing in parallel: 719 as an add-on to standard care and 451 as the anchor in dual combinations.
Both programs are expected to deliver key data in summer 2026, with SION-719 in the PreciSION CF proof-of-concept study and SION-451 in phase I dual combination studies.
The company is well-capitalized with $290 million in cash, providing runway into 2028.
Clinical strategy and decision points
The PreciSION CF study is a two-way crossover trial designed to demonstrate at least a 10 millimole sweat chloride improvement, which is expected to translate into a clinically meaningful FEV1 gain.
Patient enrollment criteria exclude those already at normal sweat chloride or with atypically high levels, focusing on those with typical responses to current therapy.
Two strategic decisions are planned for summer 2026: selecting the optimal dual combination for further development and determining whether to advance the add-on approach based on data and available capital.
Both strategies are considered commercially viable and have received positive feedback from the patient and investor communities.
