WVE-007 and body composition

• Phase I healthy-volunteer data after a single dose showed nearly 15% visceral-fat reduction, substantial subcutaneous-fat reduction, 3.3% lower waist circumference and 1% lower body weight.

• The Phase I population had no comorbidities and ~1 L of baseline visceral fat, versus 5 L in a Phase IIa comparator population; BMI alone does not capture these differences.

• INLIGHT Phase IIa is enrolling patients with BMI 35–50, with or without diabetes; four arms test two dose groups and diabetic/non-diabetic cohorts to inform dose and frequency.

• A data cut may be made at three or six months; lean mass stabilized by month three while fat continued to decline, and timing will be decided based on body-composition and weight endpoints.

INHBE development plans

• INLIGHT includes MRI-PDFF liver-fat measurement, MRI body composition, scale weight, insulin sensitivity and lipid profiles to assess obesity and broader cardiometabolic potential.

• A tirzepatide combination study is running alongside monotherapy, with potential use in patients seeking additional fat and weight reduction or individualized incretin titration.

• A maintenance-therapy study is planned to start this quarter; data across three use cases are expected to build through 2027 and inform INHBE’s cardiometabolic opportunity.

siRNA chemistry and platform

• Stereochemistry and chemical modifications improved AGO2 loading nearly 15-fold; comparisons with best-in-class TTR siRNAs showed stronger, more durable knockdown, supporting potential once-yearly dosing.

• In obese mouse models, a single dose produced weight loss; combination treatment doubled weight loss and supported maintenance of the reduction, distinguishing the construct from cited multi-dose studies.

• The platform is being extended to extrahepatic silencing as well as liver targets, using chemistry to pursue deeper and more durable suppression.
