Company presentation summary
29 Oct, 2026Platform and market opportunity
Targeted protein degradation uses E3 ligases to drive target-protein degradation; its catalytic action may enable sustained pathway blockade with lower or shorter drug exposure than conventional inhibitors.
The presentation targets historically undrugged, genetically or clinically validated pathways, aiming to pair oral convenience with biologics-like activity.
An estimated 160M patients across the US, EU5 and Japan have diagnosed immunological diseases; about 5M receive advanced systemic therapies, in markets forecast to exceed $100B in annual sales.
Advanced systemic immunology treatments are predominantly injectable biologics; cited research found over 90% of surveyed patients would switch to an oral option.
Pipeline and milestones
Wholly owned oral degrader programs include STAT6 candidate KT-621 for Type 2 diseases and IRF5 candidate KT-579 for autoimmune diseases; KT-621’s estimated patient opportunity exceeds 140M and KT-579’s exceeds 10M.
KT-621 Phase 2b AD data are expected by year-end 2026; Phase 3 AD trials are planned to start by mid-2027, while Phase 2b asthma data are expected late 2027.
KT-579’s LUMORA Phase 1b lupus trial is planned to start by early 2027; KT-485 and KT-200 are partnered IRAK4 and CDK2 degrader programs, respectively.
Sanofi initiated KT-485 Phase 1 testing in healthy volunteers and hidradenitis suppurativa patients; Gilead plans IND-enabling work on KT-200 to support an IND filing in 2027.
KT-621 clinical profile
In the 22-patient, 28-day Phase 1b AD study, median STAT6 degradation reached 98% in blood and 94% in skin across dose groups; skin degradation was maintained through treatment.
At Day 29, overall mean EASI change was -63%, EASI-50 response was 76%, and EASI-75 response was 29%; mean Peak Pruritus NRS change was -40%.
Type 2 biomarker reductions included 74% median TARC inhibition and 73% Eotaxin-3 inhibition; in AD patients with comorbid asthma, median FeNO fell 56% at Day 29, with all four patients meeting the ACQ-5 responder threshold.
The Phase 1b study reported no serious or severe adverse events, no related treatment-emergent events leading to discontinuation, and no clinically relevant vital-sign, laboratory or ECG changes.
Phase 1 results were described as in line with, or in some cases numerically above, published dupilumab Week 4 data; comparisons are cross-trial, not head-to-head, and may not be reliable.
