Vision and therapeutic approach

• Pursuing a unified approach to cancer and autoimmune diseases by targeting B-cell lineage dysfunction with in vivo CAR-T therapies.

• In vivo CAR-T aims to overcome manufacturing and patient eligibility challenges of traditional autologous CAR-T by engineering cells directly in the patient, using mRNA-LNP and lentiviral vectors.

Pipeline and partnerships

• Announced a new partnership with Senlong Biotechnology, expanding to in-license global ex-China rights for a clinical-stage lentiviral in vivo CAR-T asset.

• Two lead assets: TPST-4903 (lentiviral, dual BCMA/GPRC5D targeting) and TPST-4003 (mRNA-LNP, dual CD19/BCMA targeting), both in early clinical development.

Clinical data and efficacy

• Six patients enrolled in a dose escalation study for TPST-4903, showing promising safety and CAR-T cell expansion similar to autologous CAR-T.

• mRNA-LNP asset demonstrated 100% complete response rate and excellent safety in CAR-T naive patients, with all evaluable patients MRD negative at 12 months.

• Dual targeting CARs show superior progression-free survival in extramedullary disease compared to market leaders.
