American Society of Nephrology (ASN) Kidney Week 2026 summary
24 Oct, 2026Kidney portfolio strategy
R&D targets high-unmet-need kidney diseases with known causal biology and validated targets across AMKD, IgAN, PMN and ADPKD; programs have first- and/or best-in-class potential.
Four potential kidney launches are targeted by 2030; renal programs could exceed $13B in revenue over time.
The four programs were also framed as a potential ~$10B franchise.
ADPKD is in phase II proof of concept, initially targeting ~35,000 people, or 10% of an estimated 350,000 worldwide.
Zai Lab is positioned to market in China and other regions, while Ono will market in Japan and other regions; povetacicept is designed for monthly small-volume auto-injector dosing.
AMKD and inaxaplin
In the 13-week AMPLIFIED phase 2b trial, UACR fell 42.7% and 17.3%, while UPCR fell 44.7% and 25.4%, in the modest-proteinuria and type 2 diabetes cohorts, respectively.
AMPLIFIED was generally safe and well tolerated, with no inaxaplin-related serious adverse events; all adverse events were mild or moderate, and headache was most common at 7.3%.
AMPLITUDE completed enrollment with 522 patients, including 66 carried over from phase IIb and 456 newly enrolled; interim analysis is targeted for early 2027.
The interim analysis will assess week-48 UPCR change and eGFR slope with at least 48 weeks of data; FDA discussions on potential accelerated approval are anchored to one-year eGFR.
Management plans to discuss the type 2 diabetes cohort and potential labeling with FDA after AMPLITUDE results.
Povetacicept in IgAN
RAINIER is a global phase III trial in high-risk adults with biopsy-proven IgAN; its interim analysis assessed week-36 proteinuria, biomarkers, safety and tolerability.
At week 36, 24-hour UPCR fell 52.0% from baseline; the placebo-adjusted treatment effect was 49.8% (95% CI 39.2%–58.6%; P<0.0001), with reductions evident by week 4 and consistent across subgroups.
Gd-IgA1 fell 77.4%; hematuria resolved in 85.1% of participants with baseline hematuria, with median time to first resolution of 13 weeks.
At week 36, 42.2% achieved UPCR <0.5 g/g and 18.0% achieved <0.3 g/g, versus 6.2% and 1.5% on placebo, respectively.
Povetacicept was generally well tolerated, with serious adverse events similar to placebo; dosing is every four weeks via small-volume auto-injector. RAINIER interim data were published October 23, 2026; PDUFA is November 30.
